چکیده

The influence of divalent cations (Cd2+, Zn2+, Sr2+) and staphylococcus protein A on the superprecipitation reaction of cardiac muscle actomyosin was investigated using methods of preparative protein chemistry, optical spectroscopy and mechanokinetic analysis. It was shown that the metal ions in the range of concentrations 0.1–5 mM inhibit the Mg2+-dependent SPP reaction of actomyosin cardiac muscle and alter the kinetic parameters of this process. It was revealed that protein A modulates superprecipitation dynamics decreasing the maximal value of optical density and time of its halfmaximum achievement as well as starting and normalized superprecipitation rates. Thus, investigated factors are able to influence the actin myosin interaction changing their functional parameters of proteins from cardiomyocyte contractile complex. The kinetic characteristics of actomyosin superprecipitation are sensitive to the influence of physicochemical and pharmacological factors and can be used to study their influence on the molecular mechanisms of muscle contraction.

The role of calcium in the superprecipitation of actomyosin.

In living muscle, calcium appears to be intimately involved in the reactions leading to contraction (l-4). In isolated actomyosin systems (e.g. glycerol-extracted fibers and myofibrils) the addition of calcium will result in contraction under certain conditions: when adenosine triphosphate, magnesium, and one of several relaxing factors are present, e.g. ethylenediaminetetraacetic acid, adenosi...

The Clearing Response of Actomyosin to Adenosinetriphosphate by Samuel S. Spicer

The fact that adenosinetriphosphate (ATP) causes precipitation and contraction (superprecipitation) of actomyosin from colloidal suspension was first observed by Szent-Gyorgyi (1). It is thought that superprecipitation provides an in vitro model of muscle contraction. However, skeletal muscle contains more than enough ATP to induce actomyosin contraction in vitro (1) ; thus the question arises ...

Effect of fullerene C60 on ATPase activity and superprecipitation of skeletal muscle actomyosin.

Creation of new biocompatible nanomaterials, which can exhibit the specific biological effects, is an important complex problem that requires the use of last accomplishments of biotechnology. The effect of pristine water-soluble fullerene C60 on ATPase activity and superprecipitation reaction of rabbit skeletal muscle natural actomyosin has been revealed, namely an increase of actomyosin superp...

[On the relationship between ATPase activity and superprecipitation of actomyosin].

The relationship between ATP-ase activity and the superprecipitation of actomyosin which was extracted from the rat's uterine muscle was investigated. Results were as follows: 1) The superprecipitation occurred in the wide range of KCI concentration, 0.05-0.20 M, and particularly strongly in the range of 0.08-0.12 M. 2) In this range of KCl concentration, actomyosin-ATP ase activities were near...

Superprecipitation is a model for in vitro contraction superior to ATPase activity.

We investigated which activity of myosin B (natural actomyosin), super-precipitation or ATPase, is better suited as a parameter for measuring in vitro contraction with emphasis on the response of myosin B to Sr2+. The Sr2+ concentration giving half-maximum (KSr2+) binding of Sr2+ to cardiac troponin was not different from that to skeletal troponin, whether the troponins were contained in the ac...
